Innovative Approach from Australian Scientists Targets Triple-Negative Breast Cancer Progression

    August 22, 2026

ADELAIDE, Australia / RankWire.AI / – In the ongoing fight against aggressive breast cancers, Australian researchers affiliated with Adelaide University and the Olivia Newton-John Cancer Research Institute have pinpointed a new molecular mechanism that influences tumor metastasis. Their findings, published in EMBO Molecular Medicine, demonstrate a promising strategy for addressing triple-negative breast cancer by re-establishing a vital regulatory molecule known as miR-342. This breakthrough paves the way for novel methods to prevent lethal secondary cancers in organs such as the lungs and bones.

Australian team finds new way to tackle triple-negative breast cancer
Medical scientists analyze molecular data screens to evaluate novel targeted cancer therapies.

Triple-negative breast cancer, which makes up about 10% to 15% of Australia’s roughly 21,000 breast cancer diagnoses each year, accounts for a disproportionate share of mortality. This subtype is characterized by the absence of estrogen, progesterone, and HER2 receptors, making standard hormone-targeted treatments ineffective. Researchers revealed that when levels of miR-342 decrease, an oncogenic pathway called E2F becomes excessively active, enabling dormant cancer cells to disseminate and develop into dangerous secondary tumors throughout the body.

Biomarker Testing Could Help Identify Patients Who Would Most Benefit

Pre-clinical studies demonstrated that boosting miR-342 levels significantly limited cancer spread to distant organs. Additionally, the research showed that palbociclib, a CDK4/6 inhibitor already approved for hormone receptor-positive breast cancers, effectively suppressed metastatic tumor growth in models with low miR-342 expression. These results imply that assessing miR-342 levels could enable physicians to repurpose existing drugs for high-risk patients, offering tailored treatment options.

Associate Professor Philip Gregory from Adelaide University’s Centre for Cancer Biology, co-lead author of the study, emphasized that preventing metastasis is still the biggest hurdle in managing aggressive breast cancers. Gregory explained that because palbociclib targets the hyperactive E2F pathway, administering the medication after cancer cells have spread can prevent microscopic deposits from enlarging. Instead of merely shrinking primary tumors, this approach aims to halt the development of microscopic secondary tumors that can become life-threatening.

miR-342 as a Master Regulator of Cancer-Related Genes

The research team highlighted that triple-negative breast cancer presents significant biological variability, which has traditionally complicated the development of universal targeted therapies. By identifying a specific vulnerability common to a particular subgroup, this research opens the possibility for personalized treatment strategies. As Australian scientists advance this promising method to combat triple-negative breast cancer, efforts are underway to validate these findings using patient-derived models before moving into clinical trials.

Across Australia, medical oncologists and cancer research organizations have expressed support for these new insights, stressing the critical need for expanded treatment options when primary therapies fail. The research team intends to work with international clinical networks to speed up biomarker screening processes. Confirming the efficacy of miR-342 testing could soon enable clinicians to select patients suitable for targeted CDK4/6 inhibitor treatments early in the disease course.
